Rapid circulatiing tumor cells staining in three-dimensional microwell perfusion chip

摘要

Immunofluorescence is widespread method for circulating tumor cells (CTCs) detection and characterization. A conventional process of immunofluorescence staining on cells requires centrifugal force to wash residual fluorescence, which is not only time consuming but also cells harmful. To speed up the staining process, a three-dimensional (3D) microfluidic device with a porous PDMS membrane is employed in this work. Through a gentle perfusion process, this device can reduce staining process time from 1-1.5 hours down to 30 minutes and still keep cell in high viability.
